低溫保存對美人蝦(Stenopus hispidus)胚胎影響及冰晶形成之研究

因現今人類過度捕撈與生態之破壞,海洋物種正面臨嚴重的生存危機。美人蝦(Stenopus hispidus)是一種非常受歡迎的海洋水族館與清潔蝦的物種。擁有觀賞及貿易的經濟價值,而低溫冷凍保存美人蝦胚胎或早期幼體階段將顯著減少野生美人蝦的採集及珊瑚礁破壞並供應市場,降低對生態的影響從而提高珊瑚礁的保育。本研究的目的在了解不同發育期美人蝦胚胎對低溫、醣類、脂肪酸抗凍劑及不同海水鹽度對於胚胎孵化率的影響關係,並利用低溫顯微鏡分析不同胚胎冷凍保存過程及不同抗凍劑濃度對於胚胎顯微結變化的影響關係。研究結果顯示雖然添加抗凍劑後,可增加美人蝦的胚胎細胞在低溫下其存活率,但低溫的處理對於胚胎仍具有傷害,且隨

著胚胎在低溫下的暴露時間增長,胞器發生融合現象並產生不規則液泡,而利用抗凍劑處理後發現胚胎胞器的融合現象降低,本研究結果也顯示醣類抗凍劑在低溫處理後對於美人蝦胚胎的孵化率並無顯著提升的效果,脂肪酸抗凍劑及調整海水濃度作為抗凍劑則提升孵化率,且在脂肪酸抗凍劑及海水加入1M Meth後進一步提升美人蝦胚胎孵化率,本研究深入探討美人蝦胚胎低溫保存過程中造成死亡之機制,以做為未來最佳冷凍保存程序建立的參考。

薑黃植物是薑黃科的一種熱帶草本植物,原產於亞洲南部,在傳統上有重要的染色功能,並被廣泛地作為香料和常見的食品添加劑。而薑黃素是一種從薑黃中提取的一種黃色天然色素,具有多種藥理作用,包括消炎,抗氧化,抗細胞增生和抗血管新生等活性。薑黃酮則是薑黃油的主要成分,具有抗發炎之效果。目前薑黃在臺灣並無栽培品種的育成,薑黃相關產品因材料來源混亂,致使其產量、薑黃素含量及抗氧化活性表現等大不相同,先前的研究表明薑黃素具有良好的生物活性,但是無使用超臨界CO2萃取台灣薑黃植物方式之相關研究。因此,本研究的目的是使用超臨界CO2萃取薑黃,並確定超臨界CO2萃取薑黃的最佳條件並與其他萃取方式,使用95%酒精萃取

(EE)及索式萃取(SOX)比較分析薑黃萃取物之生物活性及抗氧化作用。此外超臨界萃取關鍵包括壓力,溫度和原料粒徑大小及輔溶劑添加。此外,我們希望發現不同方式萃取薑黃後,其萃取率、成分分析及生物活性成分是否有相似或不同之處。本研究的薑黃粉末為行政院農業委員會農業試驗所嘉義農業試驗分所提供的在地薑黃,分別利用超臨界CO2萃取(SFE),酒精萃取(EE)與索式萃取(SOX)獲得薑黃萃取物,並通過GC-MS鑑定其薑黃萃取物的主要成分,γ-himachalene,ar-turmerone,turmerone和curlone。此外,我們還利用分光光度計發現另一個成分為“薑黃素”並使用在DPPH和ABTS抗

氧化測定方法比較這幾種薑黃萃取物(SFE、EE & SOX)的抗氧化活性。總體而言,我們的結果表明,使用超臨界流體萃取(SFE)與另外兩種不同方法(EE & SOX)萃取的薑黃萃取物都具有抗氧化活性,而超臨界CO2薑黃萃取物會隨著原料的粒徑差異及輔溶劑有無添加,而有不同的生物活性及成分含量。此外,與另外兩種方法(EE & SOX)萃取的薑黃萃取物相比之下,SFE在溫度45℃,壓力 300 bar及添加95%輔溶劑 (0.5 ml/g) 時萃取率最高4.57%,薑黃色素在 SFE2- 50mesh萃取物含量為最高(1.35 mg/g);而SFE1-120mesh為最低(0.1mg/g),總多酚在

SFE2-50mesh薑黃提取物中為最高(5.19m g/g) ;而酒精萃取為最低(1.49m g/g),SFE2- 120 mesh萃取的三萜類化合物含量為最高(154.57 mg/g),但與SFE2- 50mesh (150.03 mg/g)並無明顯差異,超臨界CO2萃取的薑黃萃取物SFE的生物活性高於其他萃取方式。因此,超臨界流體萃取可能提供另一種薑黃萃取的方式。
